2 g soil was added to a flask containing 100 ml complex medium and incubated at 37°C in a shaker at 180 r.p.m. After 72 h of the incubation, enriched cultures were serially diluted up to 10-7 and 0.1 ml of diluted culture was spread over the CM agar (agar, 3 % w/v) plate and incubated at 37°C. |
