    [Ref.: #67831] Isolation/enrichment procedure 5 g pit mud was suspended in 100 ml sterile NaCl solution (0.85 %, w/v) using a 250 ml Erlenmeyer flask and mixed thoroughly. Before inoculating into liquid reinforced clostridial medium, the mud suspension was heated at 80°C for 10 min to kill the vegetative bacterial cells. After autoclaving at 121°C for 20 min, the RCM was kept in an anaerobic system overnight with a gas mixture, including 80 % N2, 10 % CO2 and 10% H2, to reduce oxygen in the medium. Ten millilitres of the mud suspension was incubated in 100 ml RCM and cultured anaerobically for 7 days at 37°C.
