Water samples were concentrated via filtration through a 0.45 µm membrane filter, with the concentrated particulate material on the filter being resuspended in approximately 3 ml of sterile lake water filtrate. Lake Water (LW) agar plates (15 g agar autoclaved in 250 ml distilled water, upon cooling 25 µg ml^-1 filter-sterilised ampicillin added and the volume made up to 1 l with the sterile filtrate that resulted above) were spread with 100 µl of the water concentrate.
