The seawater samples were quickly inoculated into a sterile fluid enrichment medium. The medium (per litre: 5 g peptone, 1 g yeast extract, 0.01 g FePO 4 , 0.3 g cysteine and 0.001 g resazurin) was prepared anaerobically and kept under an atmosphere of mixed gas (10 % H2 , 20 % CO2 and 70 % N2 ). Enrichment cultures were incubated at 16°C in 10 ml vials sealed with butyl-rubber stoppers. After 1 month, the bacterial liquid was coated on marine agar 2216 in an anaerobic packet of AneroPack-Anaero for bacterial purification. | ||
