The surface-sterilized leaf tissues of wheat (Triticum aestivum) were mixed with enrichment culture medium with 1.0 % chitin (crab shell) flakes as carbon resource and incubated at 28°C for 7 days. The chitin-enriched microbiome of the leaves was then serially diluted and coated on a plate with colloidal chitin as the sole carbon source and the plates were incubated at 28°C for 2-3 days. |
