Isolation/enrichment procedure |
The roots were surface sterilized as follows: sequential immersion in 70 % (v/v) ethanol for 5 min and sodium hypochlorite solution (4 % available chloride, freshly prepared) for 5 min. Then the sample was washed three times with sterile distilled water to remove surface sterilization agents and soaked in 10 % NaHCO3 solution for 10 min to eliminate growth of endophytic fungi. One gram of the sample was cut into small pieces with sterilized scissors and then crushed with mortar and pestle in 9 ml sterile sodium dodecyl sulfate (SDS) solution (0.1 g SDS, 1.75 g KH2PO4, 3.5 g K2HPO4, 1 l distilled water), 10-fold serially diluted in SDS solution and plated on humic acid-vitamin agar and incubated at 28°C for 21 days. |
