Isolation/enrichment procedure |
healthy plant samples were washed in water with an ultrasonic step (160 W, 20 min) to remove soil and pretreated according to the method of Lodewyckx et al.. Plant tissues were then subjected to a standard surface sterilization procedure as described by Coombs and Franco. The surface-sterilized stems were cut into pieces and crushed under aseptic conditions, serially diluted with sterilized water, followed by spreading on humic acid-vitamin agar plates. The plates were incubated at 28°C for 2-6 weeks. |
