The samples (1.0 g) were ground into powder with a pestle and suspended in 10 ml distilled water. Aliquots of serial dilutions were spread onto starch-casein agar (1.0 %soluble starch, 0.03 % casein, 0.2 % KNO 3 , 0.2 % NaCl, 0.2 % KH2PO4 , 0.002 % CaCO3 , 0.005 % MgSO47H2O, 0.001 % FeSO47H2O and 1.8 % agar; pH 7.2) containing 60 % (v/v) artificial seawater and the plate incubated at 28°C for 2 weeks. |
