Isolation/enrichment procedure |
The soil sample (5 g) was suspended in distilled water (2 ml) followed by an ultrasonic treatment (160W) for 3 min. After the addition of distilled water (43 ml), 3 % (w/v) yeast extract (1 ml) and 2.5 % (w/v) SDS (1 ml), the soil suspension was incubated at 28°C and 250 r.p.m. on a rotary shaker for 20 min. Then, 200 µl of the suspension was spread on CMKA medium supplemented with cycloheximide (50 mg l-1 ) and nalidixic acid (20 mg l-1) and incubated for 14 days of aerobic incubation at 28°C. |
